Daily intake of broccoli sprouts normalizes bowel habits in human healthy subjects

چکیده
Chronic oxidative stress impairs regular defecation. Sulforaphane (SFN) enhances anti-oxidant systems, ameliorating oxidative injury. SFN inhibits overgrowth of anaerobic microflora and protects small intestine from oxidative injury. We assessed whether daily intake of SFN-rich broccoli sprouts (BS) improves defecation in humans. Forty-eight subjects, with a constipation scoring system (CSS) >2 points, were assigned to either the BS group (n = 24) or the alfalfa sprouts (AS) group (n = 24), and were requested to eat 20 g daily of raw BS or AS, respectively, for 4 weeks. BS contains 4.4 mg/g sulforaphane glucosinolates (SGS), while AS contains no SGS. CSS-based questionnaires were performed to evaluate bowel habit. Stool samples were collected to evaluate intestinal microflora using a terminal restriction fragment length polymorphism flora analysis. Intervention with BS, but not AS, caused a significant decrease in the duration of attempted defecation and the total CSS score. Intervention with BS decreased the percentage of Bifidobacterium in the stool. These results suggest that daily intake of BS improves bowel habit in human subjects. Since BS treatment enhance antioxidant enzyme activities, these effects of BS appear to relate with the SFN-mediated modulation of the intestinal motility during exposure to oxidative stress. Chemoprotective glucosinolates and isothiocyanates of broccoli sprouts: metabolism and excretion in humans.
Broccoli sprouts are a rich source of glucosinolates and isothiocyanates that induce phase 2 detoxication enzymes, boost antioxidant status, and protect animals against chemically induced cancer. Glucosinolates are hydrolyzed by myrosinase (an enzyme found in plants and bowel microflora) to form isothiocyanates. In vivo, isothiocyanates are conjugated with glutathione and then sequentially meta...
متن کاملAbsence of the glutathione S-transferase M1 gene increases cytochrome P4501A2 activity among frequent consumers of cruciferous vegetables in a Caucasian population.
The cancer protective effect of cruciferous vegetables has been attributed to induction of phase II enzymes. But cruciferous vegetables also induce cytochrome P4501A2 (CYP1A2), which catalyzes the metabolic activation of various procarcinogens, including aromatic amines in tobacco. Thus, frequent intake of cruciferous vegetables could also result in cancer-enhancing effects. GSTM1 is involved i...
متن کاملEffects of regularly consuming dietary fibre rich soluble cocoa products on bowel habits in healthy subjects: a free-living, two-stage, randomized, crossover, single-blind intervention
BACKGROUND Dietary fibre is both preventive and therapeutic for bowel functional diseases. Soluble cocoa products are good sources of dietary fibre that may be supplemented with this dietary component. This study assessed the effects of regularly consuming two soluble cocoa products (A and B) with different non-starch polysaccharides levels (NSP, 15.1 and 22.0% w/w, respectively) on bowel habit...
متن کاملOff-target effects of sulforaphane include the derepression of long terminal repeats through histone acetylation events.
Sulforaphane is a naturally occurring isothiocyanate in cruciferous vegetables. Sulforaphane inhibits histone deacetylases, leading to the transcriptional activation of genes including tumor suppressor genes. The compound has attracted considerable attention in the chemoprevention of prostate cancer. Here we tested the hypothesis that sulforaphane is not specific for tumor suppressor genes but ...
متن کاملEnhancing sulforaphane absorption and excretion in healthy men through the combined consumption of fresh broccoli sprouts and a glucoraphanin-rich powder.
Sulforaphane (SF) is a chemopreventive isothiocyanate (ITC) derived from glucoraphanin (GRP) hydrolysis by myrosinase, a thioglucoside present in broccoli. The ability of broccoli powders sold as supplements to provide dietary SF is often of concern as many supplements contain GRP, but lack myrosinase. In a previous study, biomarkers of SF bioavailability from a powder rich in GRP, but lacking ...
